Swarming Media
- 1% (w/v) tryptone
- 0.5% (w/v) yeast extract
- 1% (w/v) NaCl
- 0.6% (w/v) Eiken agar (not regular agar!)
- 0.5% (w/v) Glucose (add from a sterile 20% glucose solution after autoclaving)
Recomendation:
Make 1500mL: 15g tryp, 7.5g yeast, 15g NaCl, 9g agar in total (do not add to batch media)
Partition into 12 bottles with 120mL1 in each (0.75g agar each)
Swimming Media
- 1% (w/v) tryptone
- 0.5% (w/v) NaCl
- 0.25% (w/v) agar
Reccomendation:
Make 1500 mL: 15g tryptone, 7.5g NaCl
Partition into 120mL bottles with 0.3g agar each
AAS Media
Same thing as swimming media but without the agar
Need approximately 10 grams of cells; from experience a total of 3L has been enough, so make 1L batches in 2L E-flasks x 3.
